An employee resigned May 27, 2008. He has just filed for unemployment benefits against my Company. How far back can he go?

You should deny this unemployment claim, and if this former employee is awarded benefits, you should appeal that decision. Wehn figuring unemployment, state agencies look at the employment record for up to 2 years prior. However, generally an employee who quits (or resigns) does not qualify for unemployment. If he has worked since May 2008, then usually that employer would be the chargable party.

We assume that you have a written resignation from the employee.

Howeve,r be aware that there is some variation in unemployment laws from state to state — and you do not mention where your business is located.
